Let's take Thor for example.
At level 1, he has 14 physical protections (without any items).
At level 20, he has 72 physical protections (without any items).
Do you see now why Titan's Bane is more situated for the late game? If you have further questions, let me know, I'm happy to discuss.

So just like inherent protections which increase per level, you'll face more protection items later.

Let's assume your guide is for arena. I typically build my 2nd item by level 8ish.
At level 8, Thor has 37 physical protections (no items).
Titan's Bane penetrates 15% of the target's defense if defense is under 65. 15% of 37 is 6 penetration.
Let's assume, for whatever reason, Thor builds Breastplate of Valor 2nd item at level 8. (gives 65 protections). He now has a total of 102 protections.
If enemy has over 65 protections, the penetration formula for Titan's Bane is:
(((protections-65)/135)*25)+15
You now have 22 penetration against Thor, which is more than most flat penetration items.
But here's what you have to consider. Camazotz is an assassin. In the early game, he's gonna want to dive a squishy enemy (mage/hunter), burst him down, then get out. You shouldn't be focusing tanky targets that early unless they're low on health or you have enough backup to burst him down. That's why those flat penetration items would take precedence over Titan's Bane in the early game.
You're free to choose your own playstyle and build however you want though.
But not only is this NOT a static % pen value (like Exe), it's a SCALING % pen, which makes it even MORE of a difference the higher the protections.
You can infer this concept without having to be extremely precise in your wording. Hopefully you're getting the concept of what we're trying to explain, meme.
So in general, % pen / protection reduction is better NOT being built early. But you can go further. Static % is going to be better earlier than scaling %. This is also why you see The Executioner and Demonic Grip built mid-game (3rd-4th slot sometimes), while Titan's Bane and Obsidian Shard are typically seen in the 5th or later slots.
Look, the proper build order would be something like: Warrior Tabi, The Crusher, Jotunn's Wrath, Void Shield, Heartseeker, Titan's Bane.
