Thoth conjures three Hieroglyphs around him, ignoring the Basic Attack Movement penalty. His next 3 Basic Attacks pass through Enemies and apply Ability Damage instead. Additionally, these Basic Attacks deal an additional 25% damage to jungle minions. If these attacks pass through the Glyph of Pain, their range is increased by 30. Hitting an enemy with this ability increases the damage they take from each subsequent shot by 10% and reduces the cooldown by 0.5s (this can only occur once per shot).

This projectile roots the first Enemy god hit while slowing all other enemies, and also deals Ability Damage.

If either the Dash or Attack passes through the Glyph of Pain, their respective range is increased by 30.

From his book, Thoth conjures forth a Hieroglyph in front of him. Thoth's ability projectile passing through the glyph receive increased damage, and a range increase of 30. This damage does not trigger item effects.

Thoth and allies' Basic Attack projectiles also receive a lesser damage bonus, but no range increase.

Thoth begins summoning pages from his book, up to a maximum of four. When all four have been summoned, he sends them forward damaging all Enemies in their path.

Thoth may activate this ability again to fire the ability early, doing a reduced amount of damage based on the number of glyphs currently summoned. Alternately he may cancel this ability at a cost of 30 Mana.

Intro

From my experience so far with Thoth, from what I've seen, his three is crucial. Aim to shoot abilities and basics through it if at all possible (and sensible). His dash has a long cooldown, so I recommend playing him as if he didn't have one the majority of the time. His one is his main clear, but doesn't seem to do enough without his three. Combo-ing his three and his ultimate means you can do colossal damage to single targets or in team fights.

Abilities and Combos

I shall begin by explaining his abilities.

Passive: "Thoth pens the names of his fallen opponents in his book, recording their weaknesses. After killing 30 enemies, Thoth gains a stack of 4 Magical Penetration. He unlocks a new stack every 4 levels, up to a maximum of 5 stacks at level 17."

My understanding of this is that for every thirty enemies Thoth kills (including minions and gods, not sure about jungle camps), he gains 4 penetration. Every 4 levels Thoth gains the ability to gain another stack of this, up to a max of 5 stacks of 4 penetration at level 17.

1: Thoth can fire three more powerful projectiles. These pass through enemies, doing damage to all hit. This can be interspersed by using his second, and has its range and damage increased by his third.

2: Thoth dashes in the direction he is currently travelling, and his next basic attack after doing this does additional damage. The dash distance can be increased by his 3, and the damage, but not distance, of his follow-up basic can be increased by this.

3: Thoth places a glyph that causes basic attacks by his team to do increased damage when they pass through it. This interacts with all his other abilities. See them to find out more.

4: Thoth has an Ultimate that needs to charge. It can be effected by CC and he can stop casting, without releasing the ability, with no additional cooldown. The longer Thoth charges this Ultimate, the more damage it does when activated. This ability does additional damage and gains additional range when it passes through his 3.

Combos:

The 3 is crucial to most combos, the main one being place 3, use 1 through it.
Other combos are:
3, 2 through it, used to chase or escape
3, 4 through it, 1 through it, doing tons of damage onto groups of enemies
3, 2 through it, turn, basic through it, 1 through it
3, 4 through, 2 through to chase, basic attack, 3, 1 through it
In other words, there are tons of combos you can do with Thoth.

Game Mode Tactics

Siege
With Thoth in any game mode, positioning is crucial. In siege, you normally need a very tanky character on your lane to distract the enemies' attention away from you, while you deliver tonnes of damage. Later game you may be able to take the siege juggernaut, providing you have someone to take the damage. Be very careful in the jungle in this game mode, as you have low mobility and are prone to ambushes.

Conquest
You should be playing middle lane in this game mode, as you have high damage and need solo farm. Your lane clear is good so you should be able to pressure out most other middle laners. Make sure you rotate when needed, and be very careful in the jungle.

Joust
Your lane clear potential with your 3-1 combo should make you hard to deal with in this game mode. Taking damage camp can make you a huge threat, but be sure to not over commit. You are liable to die if attacked from behind, so keep an eye out.

Clash
Positioning is especially key here. Switch lanes often, try and get as much solo farm as possible. Gain additional XP by taking down the furies' XP camp. As you have such high damage you will be key to your team's potential success.

Arena
In arena as Thoth, do not get flanked. If an enemy is behind you, you become incredibly vulnerable and can be killed easily. Use your large amount of clear to prevent minions entering your portal, and do not forget the power of the long range ultimate.

God Companions

ADC
This role has many possible combinations for playing if you have a Thoth on your team. If you have a hunter with a basic attack steroid, they will be a perfect combination with Thoth.

Support
Any support with high CC will benefit a Thoth. This will allow you to get off all your damage without them retaliating. Therefore the following are recommended:

Solo
As your solo you want someone who can really distract from you, and be really annoying to the other team. CC is also good.

Examples:
Bellona
Hercules
Tyr
Odin
Ravana

Jungler
You want Junglers that can finish off kills you can't quite secure, CC is good and high single target damage is recommended.

Examples:
Arachne
Loki
Mercury
Thanatos
Thor
Ratatoskr
Serquet

I have added an "Alternative Builds" section, where I have created (and borrowed) a few other builds using the items I suggested.

I haven't played Thoth yet (I'm an Xbox One Player) so, he just came out today. I get why you put Polynomicon in your build, it caters to Evade and Punish which has a projectile basic that slows much like Rama's Rolling Assault.

Personally, as a Joust player, I think i'd be keeping my distance with Thoth holding down the back-line. My standard build would probably look something like this...

Book of Thoth
Shoes of the Magi
Chronos' Pendant
Spear of the Magus
Rod of Tahuti
Gem of Isolation

This way I can have +25 Flat Penetration opposed to 30% CDR. I think the 20% CDR is going to be just fine, but I'll have to wait and see once I get my hands on him!

Thanks for the info, I hope it is all right that I have included your preferred build in the "Alternative Builds" section of my guide.
I personally like the CDR so I can have my 1 available more often, allowing you more consistent damage. Also, his passive does have some built in penetration.
